Sun exposure and cutaneous HPV infection found synergistic in skin cancers

Researchers at Moffitt Cancer Center and colleagues at the University of South Florida and the German Cancer Research Center in Heidelberg have found that having antibodies for cutaneous types of human papillomavirus (HPV), coupled with sun exposure (ultraviolet radiation) or poor tanning ability, can act "synergistically" in the development of non-melanoma skin cancers such as basal cell carcinoma (BCC) and squamous cell carcinoma (SCC).

A number of studies into the relationship between cutaneous HPV and sun exposure have been conducted previously but with mixed results, the researchers said.

This study, the first to investigate interaction effects between genus-specific cutaneous HPV positivity and multiple measures of sunlight exposure as related to BCC and SCC in a U.S. population, was published in a recent issue of The Journal of Infectious Diseases.

"UV radiation exposure is the most important risk factor for the development of non-melanoma skins cancer," said study lead author Dana E. Rollison, Ph.D., Moffitt associate member, vice president and chief health information officer. "Cases of non-melanoma skin cancers are increasing despite the increased use of sunscreen products. Thus, so that new interventions can be developed, there is a need to identify co-factors that may interact with UV radiation exposure in increasing the skin cancer risk."

According to the authors, the risk factors for basal cell and squamous cell carcinomas are male sex; age; light skin, eyes and hair; and UV radiation exposure.

UV radiation exposure and light skin pigmentation are the most recognized risk factors. People with low melanin production tend to have difficulty tanning when exposed to UV radiation.

Skin pigmentation, created by chemical melanin production in the skin, is the "main photoprotective mechanism in the skin," noted the researchers.

The researchers hypothesized that persistent HPV infection may promote tumor progression by interfering with an individual's response to UV radiation-induced DNA damage and that HPV plays a synergistic role in the development of BCC and SCC. Accordingly, their goal was to investigate the potential "modifying effects of cutaneous HPV seroreactivity on the associations between sunlight exposure, host susceptibility to UV radiation exposure, and both BCC and SCC."

The study recruited 204 patient volunteers with BCC, 156 with SCC, and 297 controls with no reported cancer types. The three groups were surveyed on demographics, personal constitutional characteristics, lifestyle factors and measurements of sunlight exposure. Each volunteer provided a blood sample for cutaneous HPV antibody measurement.

"Sun-related factors were associated with BCC and SCC," Rollison said. "Cutaneous sensitivity to sunlight exposure - specifically experiencing a blistering sunburn - and poor tanning ability were associated with a higher prevalence of antibodies to cutaneous HPV types in genus beta. The associations between poor tanning ability and SCC were significantly greater among those positive for antibodies to cutaneous HPV types in genera alpha and beta."

Additional studies are needed, including those measuring infection with cutaneous HPV types in multiple genera, concluded the researchers.

"Identifying how HPV infections might influence sunlight-associated risks of NMSC may lead to improved identification of high-risk individuals and also aid in the development of new prevention strategies," Rollison said.

Family Home and Life: The Fun at Gramma's House

What does red and green nail polish on my kitchen counters, candy sprinkles on the kitchen floor, and an over tired Gramma and Papa have in common? A pajama party at Gramma?s house of course!

It was our first ever over night with the grand kids at our house. We have stayed at the kids houses over night before but to have them at our house was a real treat. We have a scorpion problem in our neighborhood, and our house had an infestation of scorpions when we moved in. Scorpions are most active at night.?It was not uncommon to find several scorpions a day in the house. One night we found eight in about 15 minutes. I thought I was going to lose my mind over those creepy things! ?I have found them in kitchen drawers, under my dishwasher handle, in my dryer lint catcher, on the walls and ceilings as well as the usual dark places you might expect to find them. I have unknowingly showered with them and even had one crawl up my leg while I was sitting at my sewing machine. Yes, I have been stung, on my arm one night while I slept. ?I am so thankful a child has never been stung! It took a few years to get that problem under control and we rarely see one now.

Everyone felt it would be ok to have the kids over night, so we had the five older girls come and stay. We started the night off with letting them just run around and have fun. They were so excited to be having a PJ party together. It was loud in my house, lots of screaming and giggling going on. Then dinner and then it was makeup time. One of them had seen an episode of Maxx and Ruby and the girls there had ?make overs?.? They also brought nail polish??even though I had them stay in the kitchen trying to keep it all under control I still ended up with it on my counter. It came off though so no worries. Next time no nail polish I think.

Then they wanted to dance so I put a kid?s music cd in the player and let them get rid of more energy. Now it was time for ice cream and other sugary things.? After, I sent them to the bathroom to brush their teeth and we all crowded (except Papa, he was?M.I.A. by then) into my big bed for a movie. The two smaller girls would later sleep in pac n plays I set up at the end of my bed. (no one would ever dream of sleeping on the floor here because of the scorpions) The plan was to move them to the pac?n plays?when they feel asleep. I realized that was not going to happen and eventually put them in the pac n plays and they were asleep in seconds! The older two stayed awake for a while full of giggles and wiggles but finally all were asleep and I got as comfortable as I could on the edge of the bed and slept too. Papa slept peacfully and uneventfully on the new bed we recently put in the playroom.

The party?was fun and I do want to have one again; I learned a few things about PJ parties too.?It won?t be noticed if you vacuum and mop before they kids come. Save yourself the effort because you will do it after they leave anyway. ?Asking them to brush their teeth before going to bed can be premature as they may all decide they want another snack just after climbing under the covers and will need to brush again. Anything and everything is fodder for giggles and silliness with a girls PJ party.? The next day you will be very tired?so plan for that and don't have to go to work like Papa and?I did......waaaahh!!!!

Publication of flu study reveals full nature of threat

It's one of the most troubled scientific gestations on record. Nine months after it was first presented at a conference, research showing that just five mutations lead to a deadly H5N1 bird flu which transmits through the air between mammals has finally been published. Estimates published with it show that nature might well produce this virus too ? and reveal the science we now need to head off that threat.

The research, by Ron Fouchier and colleagues at Erasmus Medical Centre in Rotterdam, the Netherlands, was presented at a meeting and reported by New Scientist last September. But its publication ? and that of similar work by Yoshi Kawaoka at the University of Wisconsin-Madison ? was delayed by concerns that other labs would recreate airborne H5N1 with bioterrorist intent, or that insufficient containment might allow the virus to escape. In March, an advisory committee of the US agency which funded the work voted that these risks were outweighed by the benefits of publishing.

The main benefit was that it alerts us to some of the warning signs that might suggest one of the vast number of H5N1 viruses circulating in birds could become a pandemic. Now that both studies have been published, it is clear why they felt this would be a good idea.

Each lab did different things to H5N1 to make it transmissible. All but one of the five mutations that did the trick were different. Yet all did remarkably similar things to the virus. "Now we know what changes in the behaviour of the virus can make it transmissible, we can watch for any mutations with those effects ? not just our particular ones," says Fouchier.

For one thing, you need to change the HA surface protein so it binds a cell-surface sugar in mammals' noses, instead of the one it binds in birds. You also need a mutation in the RNA-replicating polymerase enzyme that adapts it to mammals' cooler bodies.

In both studies, further exposure of these viruses to ferrets, the best experimental mammal, induced further changes. Both viruses got rid of a sugar on the tip of HA. And both turned up a further, novel mutation. Kawaoka's stabilises the virus while attacking the cell. Fouchier's is at a spot in the HA protein where, he thinks, it may have similar effects.

Beyond the lab

Can this happen in nature? In a companion paper, Derek Smith and Colin Russell at the University of Cambridge, and colleagues, say all these mutations are already seen in bird flu ? although two of the three sugar-binding mutations were in H2 or H3 viruses, not H5N1.

The polymerase adaptation, meanwhile, occurs in nearly 30 per cent of H5N1 sequenced so far. The sugar loss occurs in more than half of sequences. Both stabilising mutations have been seen in Chinese H5N1.

Russell's team calculate that in many sequenced viruses containing some of the required mutations, only three to four single nucleotides in the viral DNA must mutate to get the rest they need to go airborne.

We don't know how fast the mutations accumulate. Using a mathematical model, however, Russell's team found that a virus that needs only three more mutations could well emerge within the five-day course of a single mammalian infection.

And it could be happening already. The virus you sneeze out is a cloud of slightly different mutants. The sequences on record for H5N1 are mostly "consensus" ? an average. The rest of the nucleotide changes required for H5N1 to go rogue might already be hiding within this consensus.

"We need to get those samples out of the freezer again and do some deep sequencing to see what minority mutations are there, and how often they appear," says Smith. We also need to establish just how deadly the transmissible viruses really are. For now, such research is blocked under a moratorium on work that boosts H5N1 virulence or transmission.

Chesapeake Energy To Layoff Workers After Cutting Drilling Projects (CHK)

6-19-2012- Chesapeake Energy Corporation(NYSE:CHK) announced that they are going to cut 70 jobs as result of shutting down drilling projects in northern Texas. The move comes in the wake of severe natural gas price declines and as many competitors make similar move to offset narrowing margins. There will also be layoffs at their Fort Worth and Cleburne headquarters, in areas such as public affairs, marketing communication, administrative services, and community relations.

Chesapeake Energy Corporation produces oil and natural gas. The Company's operations are focused on discovering, developing and acquiring conventional and unconventional natural gas reserves onshore in the United States.

Chesapeake stock is currently trading at $18.71 per share, which is currently above its 50-day moving average of $17.27.

Mass. nuclear plant, union, reach contract deal

PLYMOUTH, Mass.?Management at the Pilgrim nuclear plant in Plymouth and unionized workers have reached a tentative contract agreement.

The Utility Workers Union of America Local 369 announced the agreement early Tuesday morning. Union membership is scheduled to vote on the proposal Wednesday.

Union president Dan Hurley said in a statement that the contract "has important protections" for the workers.

Entergy executive Robert Smith called the four-year labor contract "fair and equitable."

No details were released.

Contract negotiations had stalled over health care costs and working conditions.

The dispute became contentious with plant owner Entergy Corp. locking out union workers and the union filing labor complaints with the National Labor Relations Board.

In response to the agreement, the union said a rally outside the Statehouse planned for Tuesday had been cancelled.

Black Plague Leaves Oregon Man in Critical Condition

An Oregon man is in critical condition this week after apparently contracting the plague. Yes, the plague generally associated with the Middle Ages.

Oregon health officials believe the hospitalized man was infected with the "black death" plague, a bacterial infection that affects the blood stream.

According to The Oregonian, the Bend resident became ill with the bubonic plague - the cause of the "black death" - after he was bitten on the hand.

A stray cat bit him while he was trying to get a mouse out of its mouth ... for reasons unknown. He was listed in critical condition at the hospital.

It's actually the fifth case in Oregon since 1995. The stray cat has since died, and has been sent to the Center for Disease Control for testing.

Often transmitted by fleas that are infected with the Yersinia pestis bacteria, black plague is transmitted through bites, according to the CDC.

Symptoms include a swollen lymph gland known as a "bubo," fever, chills, headache, and exhaustion. Illness usually occurs 2-6 days after infection.

During the "Black Death" period starting in the late 1340s and lasting for centuries, 25 million lives were claimed, according to National Geographic.

For reference, Europe's population was around 70 million at the time.

"This can be a serious illness," said Emilio DeBess, Oregon's public health veterinarian said. "But it is treatable with antibiotics, and it's preventable."

Treatment consists of hospitalization, antibiotics and medical isolation. Only 1 in 7 cases in the U.S. end in death nowadays ... let's hear it for science!

CAIRO (AP) ? Egypt's Muslim Brotherhood is warning that a court ruling to dissolve the Islamist-led parliament and let Hosni Mubarak's former prime minister run for president is a move toward reversing the gains of the revolution.

The group, which held the most seats in parliament, says the decision will lead to more dangerous days than those under Mubarak's repressive regime.

Friday's statement says progress made since Mubarak was ousted last year is being "wiped out and overturned."

The Supreme Constitutional Court's decision leaves Egypt with no parliament and concentrates power even more firmly in the hands of the generals who took over from Mubarak.

Ahmed Shafiq, who was Mubarak's last prime minister, is set to face the Brotherhood's candidate Mohammed Morsi in a presidential run-off Saturday and Sunday.
